10x Genomics Chromium Single-Cell Gene Expression Controller

The Chromium Controller is keystone piece of equipment responsible for single-cell encapsulation. A variety of reagent kits are available for this system, including:

Miseq and Novaseq

We have one Illumina Miseq on-site—capable of up to 25 million reads with V3 reagent kits—that is useful for pilot or diagnostic runs. Large-scale Novaseq runs are outsourced to an off-campus facility. Since many users will not have enough libraries to fill an entire Novaseq Lane, we maintain a queue of samples and pool samples across groups for every Novaseq run. An automated DNA size selection technology, the BluePippin can isolate fragments ranging from 100bp to 50kb, depending on the gel cassette used.

Bio-Rad C1000 Touch Thermocycler

The Thermocycler is a key tool during single-cell library construction. SPEC has two thermocyclers on-site, one with a single-chamber for 100uL reactions and another with dual-chamber for 50uL reactions.

Countess II FL Automated Cell Counter

An automated cell counter which utilizes disposable sample slides, the Countess II FL provides cell count and viability through Trypan blue staining. The technology is typically accurate if the cell concentration is between 100,000 to 3,000,000 cells/mL.

NanoDrop One

Primarily used to verify oligo molarity, the NanoDrop One quantifies the amount of nucleic acids and sample purity via absorption.

BSL2 Cell Culture Facility

Located in BBB room 151A, our facility includes two biosafety cabinets, two CO2 incubators, a centrifuge, a 37C waterbath, a fridge/freezer, and a fluorescent microscope.
